Hubert, Earl G. (Veterans Administration Center, Los Angeles, Calif.), George M. Kalmanson, and Lucien B. Guze. Preservation of antigen-coated sheep erythrocytes by freezing for use in indirect hemagglutination procedure. J. Bacteriol. 86:569-572. 1963.-Preservation of sheep erythrocytes, coated with various protein and lipopolysaccharide antigens, was accomplished by rapid freezing and storage at -70 C in a dextrose-lactose solution. The stored cells were tested at intervals in an indirect hemagglutination procedure. Sensitivity of these cells was maintained, and antibody titers were consistently reproducible throughout the period of testing. These observations suggest that sheep erythrocytes may be coated with a variety of antigens and may be employed satisfactorily in an indirect hemagglutination test, even after storage at -70 C for a period up to 6 months.

Improvement of the indirect hemagglutination assay for Salmonella typhi Vi antibodies by use of glutaraldehyde-fixed erythrocytes.

Glutaraldehyde-fixed sheep erythrocytes sensitized with Vi antigen were shown to be usable for at least 1 year in the indirect hemagglutination assay for Vi antibodies. The use of fixed sensitized erythrocytes improves the practicality of the indirect hemagglutination assay by (i) eliminating the need to sensitize cells each time the test is performed; (ii) reducing waste of the purified Vi ant...

Indirect hemagglutination test that uses glutaraldehyde-fixed sheep erythrocytes sensitized with extract antigens for detection of Pasteurella antibody.

Glutaraldehyde-fixed sheep erythrocytes (GA-SRBC) were used in the indirect hemagglutination test for the detection of Pasteurella antibody. GA-SRBC were stable for at least 6 months. Heat extract or potassium thiocyanate extract antigens of Pasteurella strains could be adsorbed onto GA-SRBC or tanned GA-SRBC, respectively. The indirect hemagglutination test reaction was capsular group specific...

Coagglutination test for identification of Pasteurella multocida associated with hemorrhagic septicemia.

Two serogroups (B and E) of Pasteurella multocida associated with hemorrhagic septicemia in cattle were differentiated by a coagglutination test with antibody-coated staphylococci. The group antigen was soluble and heat stable. It was not detectable in unencapsulated cells or their heated extracts. Adsorbed onto human type O erythrocytes, it could be demonstrated by an indirect hemagglutination...

Micro-indirect hemagglutination test for detection of antibody against transmissible gastroenteritis virus of pigs.

A micro-indirect hemagglutination (IHA) test was developed for detecting antibody against transmissible gastroenteritis (TGE) virus of pigs. TGE virus propagated in swine kidney cell cultures was highly purified and concentrated by the combination of ammonium sulfate precipitation, treatment with fluorocarbon, and sucrose density gradient centrifugation. Tanned sheep erythrocytes were sensitize...

Evaluation of a hemagglutination test for human leptospirosis.

An indirect hemagglutination test for the diagnosis of leptospirosis is described; the test uses a soluble antigen from serotype patoc to sensitize sheep erythrocytes which are then fixed with glutaraldehyde. Evaluation of this procedure indicates that it is more reliable than the conventional macroscopic agglutination test and, in contrast with both microscopic and macroscopic agglutination te...
